Double Agents and Secret Agents: Developments of new MRI contrast agents for molecular imaging

Many MRI contrast agents have been developed during the last 20 years that can detect a variety of biomarkers. However, the specificity for detecting the intended biomarker can be challenging, and the accuracy of quantitative measurements with these MRI agents often needs improvement. One approach for addressing these problems is to use "double agents", or agents that produce two detectable signals that can be used to isolate the effect of the intended biomarker. These double agents that inherently provide more information can also provide opportunities to improve the accuracy of quantitative imaging. Examples of CEST MRI agents and PET/MRI agents will be used to highlight the advantages of double agents. In addition, an example of a T2-exchange agent will be presented, because the T2-exchange mechanism is not well-known, and therefore this agent can be considered a "secret agent" within the MRI contrast agent field.
