1) Kompartimentierung und Konnektivität des Thalamus: fMRT "Resting State" und DWI Untersuchungen

Ziel des Projeks ist es, mittels funktioneller Magnetresonanztomografie (fMRT) im Ruhezustand (rsfMRT) und diffusions­gewichteter Bildgebung (DWI) die funktionelle und strukturelle Untergliederung des Thalamus und seiner thalamo-kortikalen Konnektivitäten zu untersuchen. Zu diesem Zwecke soll erstens eine Kartierung des mensch­lichen Thalamus entwickelt und bereitgestellt werden, die für die Analyse und Interpretation bildgebender Befunde in den Neurowissenschaften verwendet wer­den kann. Diese Kompartimentierung sollen zweitens mittels Fasertraktografie (FT) auf ihre Konnektivität zu kortikalen Strukturen untersucht werden. Hierbei wollen wir auch hemi­sphärische und geschlechtsspezifische Differenzierungen erfassen. In einem dritten Schritt sol­len schließlich die funktionellen und strukturellen Segmentierungen des Thalamus mit den vorhan­denen anatomischen Atlanten in Beziehung gesetzt werden. Insgesamt hoffen wir mit diesem Pro­jekt, die Rolle des Thalamus im Konzert mit den sensorischen und kognitiven Funktionen des Gehirns genauer zu erfassen.

2) Kompartimentierung der Basalganglien: Funktionelle MRT und DWI Untersuchungen

Ziel des Projects ist es, in Fortsetzung zu den bisherigen Untersuchungen des Thalamus die relativ uniforme Anatomie der BG zu verfeinern, und eine strukturelle und funktionelle Untergliederung der drei großen Kerngebiete, des Nucleus caudatus (NC), des Putamen (PU) und des Globus pallidum (GP) zu erstellen, und deren kortikale und subkortikale Verbindungen zu analysieren. Dazu wollen wir a) die strukturelle und funktionelle Parzellierung dieser BG Kerne an zwei großen Kollektiven (3 und 7 Tesla) des Human Connectome Projects (HCP) analysieren und b) die funktionelle Untergliederung durch hochaufgelöste Bildgebung (< 1 mm3) von gesunden Kontrollpersonen am 9,4 Tesla Scanner ergänzen. Die Auswertung dieser Untersuchungen wird funktionell mit einer verbesserten ICA Methodik und strukturell mittels Fasertraktografie (FT) erfolgen. Hierbei sollen auch hemisphärische und geschlechts-spezifische Differenzierungen erfasst werden. In den bisherigen Arbeiten konnte bereits gezeigt werden, dass a) eine funktionelle Untergliederung von BG und Thalamus anhand kortikaler rsfMRT-Netzwerke (cRSN) möglich ist, und dass b) mittels DWI intrathalamische und subkortikale Verbindungen erfasst werden können.
